Production Procedures
Purified normal human IgG is isolated using fractionation and DEAE chromatography. Fc fragments were produced by papain digestion and affinity chromatography.

Protein concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

By immunoelectrophoresis the IgG was shown to 1) react with antiserum specific for human IgG-Fc, 2) not react with antiserum specific for IgA, IgM, kappa or lambda chain and 3) produce a single precipitin arc with antiserum against human serum identical to that produced with anti-IgG-Fc.

By ELISA the IgG contains less than 1% of the other immunoglobulin classes and light chains kappa and lambda.

IgG is the primary immunoglobulin protein found in blood/plasma, and functions to neutralize toxins, active complement, immobilizing pathogens and inducing opsonization, and cell-mediated cytotoxicity. The anti-Fc activity ensures activity only to the Fc portion of the IgG molecule and not the Fab fragments on the light chain.

Applications

Used as a positive control in immunoassays.

Serum or plasma from donor has been tested and found negative for HIV-1 antigen, antibodies for HIV and HCV, and non-reactive for HBsAg, HIV-1 RNA, and HCV RNA. Not tested for AntiHBc. HANDLE AS IF CAPABLE OF TRANSMITTING INFECTIOUS AGENTS. Used as a positive control in immunoassays.
