Antibody was affinity purified using an epitope specific to ACC1/ACC-alpha immobilized on solid support.

The epitope recognized by A300-986A-T maps to a region between residue 1925 and 1975 of human acetyl-Coenzyme A carboxylase alpha using the numbering given in entry NP_942131.1 (GeneID 31).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

ACC1/ACC-alpha catalyzes the carboxylation of acetyl-CoA to malonyl-CoA in the enzymatic pathway of long-chain fatty-acid synthesis. ACC1/ACC-alpha has been identified as a partner of BRCA1. Through this interaction, ACC1/ACC-alpha may contribute to the tumor suppressor activity of BRCA1 as demonstrated by the finding that disruption of this interaction increases lipid synthesis in mammary epithelial cells. In this context, lipogenesis has been closely linked to tumorigenesis and ACC1/ACC-alpha has been found to be highly expressed in human breast carcinomas.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
