Antibody was affinity purified using an epitope specific to BCR immobilized on solid support.

The epitope recognized by A302-057A maps to a region between residue 1221 and 1271 of human breakpoint cluster region using the numbering given in entry NP_004318.3 (GeneID 613).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

BCR (breakpoint cluster region) was identified as part of a fusion protein that results from a reciprocal translocation with the ABL gene between chromosomes 22 and 9. The BCR-ABL fusion is the cause of chronic myeloid leukemia (CML). The BCR protein contains a C2 domain, a DBL-homology domain, a PH domain, and a Rho-GAP domain and functions as a GTPase-activating protein for RAC1 and CDC42.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (Cat. No. A120-101P).

Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
