The epitope recognized by A303-458A maps to a region between residue 300 and 350 of human Chaperonin containing TCP1, Subunit 3 using the numbering given in entry NP_005989.3 (GeneID 7203).
Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4. Antibody was affinity purified using an epitope specific to CCT3 immobilized on solid support.

CCT3 is also known as T complex protein 1 subunit gamma (TCP1-gamma) and is a molecular chaperone that assists in the folding of proteins upon ATP hydrolysis. As part of the BBS/CCT complex, CCT3 may play a role in the assembly of BBSome, a complex involved in ciliogenesis and regulating the transport of vesicles to the cilia. CCT3 is known to play a role, in vitro, in the folding of actin and tubulin [taken from the Universal Protein Resource (UniProt) www.uniprot.org/uniprot/ P49368].

Applications
All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (Cat. No. A120-101P).
Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
