Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4. Antibody was affinity purified using an epitope specific to CPSF160 immobilized on solid support.

CPSF160/CPSF1 (cleavage and polyadenylation specific factor 1, 160 kDa subunit) is a subunit of the cleavage and polyadenylation specificity factor (CPSF) involved in mammalian mRNA 3’-end processing. CPSF, along with CstF (cleavage and stimulation factor), is critical for the recognition of cis-acting sequence elements on the pre-mRNA that dictate the site of cleavage and subsequent polyadenylation. CPSF exists as a complex of four proteins that includes CPSF160, CPSF100, CPSF73, and CPSF30. CPSF160 is responsible for recognizing the (AAUAAA) poly (A) signal for cleavage and polyadenylation.

Applications
All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (Cat. No. A120-101P).
Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
