The epitope recognized by A301-537A maps to a region between residue 991 and 1041 of human DEAH (Asp-Glu-Ala-His) box polypeptide 16 using the numbering given in entry NP_003578.1 (GeneID 8449).
Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4. Antibody was affinity purified using an epitope specific to DHX16 immobilized on solid support.

DHX16 is a member of the DEAH-box family of proteins that possesses several conserved motifs which include the highly conserved DEAH (Asp-Glu-Ala-His) amino acid sequence motif. The major activity of DEAH-box proteins is to function as ATP-dependent RNA helicases. RNA helicases play an important role in all aspects of RNA metabolism and function which include pre-mRNA splicing, RNA synthesis, RNA degradation, RNA export, RNA translation, RNA secondary structure formation, ribosome biogenesis, and the assembly of RNP complexes. DH16 was originally identified as DBP2 and discovered to be a functional homolog of the yeast RNA helicase and ATPase, Cdc28/Prp68. DHX16 appears to play a key role in nonsense-mediated mRNA decay (NMD) as well as rRNA processing.

Applications
All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
