Factor inhibiting HIF-1 (FIH-1) hydroxylates HIF-1 alpha at 'Asp-803' in the C-terminal transactivation domain (CAD). FIH-1 functions as an oxygen sensor, and under normoxic conditions, the hydroxylation prevents interaction of HIF-1 with transcriptional coactivators including Cbp/p300-interacting transactivator. FIH-1 is involved in transcriptional repression through interaction with HIF1A, VHL and histone deacetylases [taken from the Universal Protein Resource (UniProt) www.uniprot.org/uniprot/Q9NWT6].

Applications
All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
