Antibody was affinity purified using an epitope specific to MDC1immobilized on solid support.

The epitope recognized by A300-052A maps to a region between residues 925 and 975 of human Mediator of DNA Damage Checkpoint 1 using the numbering given in entry NP_055456.1 (GeneID 9656).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

Mediator of DNA damage checkpoint protein 1 (Mdc1) is required for checkpoint mediated cell cycle arrest in response to DNA damage within both the S phase and G2/M phases of the cell cycle. It may serve as a scaffold for the recruitment of DNA repair and signal transduction proteins to discrete foci of DNA damage marked by 'Ser-139' phosphorylation of histone H2AFX. It is also required for downstream events subsequent to the recruitment of these proteins. These include phosphorylation and activation of the ATM, CHEK1 and CHEK2 kinases, and stabilization of TP53 and apoptosis [taken from the Universal Protein Resource (UniProt) www.uniprot.org/uniprot/Q5PSV9].

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
