Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4. Antibody was affinity purified using an epitope specific to PARC/H7-AP1 immobilized on solid support.

PARC is a member of the cullin family of proteins that act as scaffolds to bring together ubiquitin E3 ligases, E2 conjugating enzyme, and substrate proteins to mediate the ubiquitination of substrate proteins. PARC is highly homologous to and can form heterodimers with CUL7 (cullin 7). PARC/H7 interacts with p53 and was initially thought to have a role in the sequestration of p53 to the cytoplasm. Recent studies demonstrate that PARC may not influence p53 function or localization, but rather p53 may serve to regulate PARC/H7-AP1 function.

Applications
All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (Cat. No. A120-101P).
Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
