The epitope recognized by A300-661A maps to a region between residues 1275 and the C-terminus (residue 1323) of human PH Domain and Leucine Rich Repeat Protein Phosphatase-like using the numbering given in entry NP_055835.2 (GeneID 23035).
Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4. Antibody was affinity purified using an epitope specific to PHLPPL immobilized on solid support.

PHLPPL (PH domain leucine-rich repeat-containing protein phosphatase-like) is responsible for the dephosphorylation and subsequent inactivation of Akt. Akt is a signaling protein critical to the balance of cell survival and apoptosis. Dephosphorylation of Akt by PHLPPL has been demonstrated to trigger apoptosis and suppress tumor cell growth. PHLPPL is also known as PHLPPL2 and functions similarly to its relative, PHLPP1. Recent studies show that PHLPPL and PHLPP selectively terminate Akt-signaling by targeting specific Akt isoforms.

Applications
All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (Cat. No. A120-101P).
Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
