Production Procedures
Antibody was affinity purified using an epitope specific to Roquin immobilized on solid support.

The epitope recognized by A300-514A-T maps to a region between residues 500 and 550 of human Ring Finger and CCCH-type finder domains 1 using the numbering given in entry NP_742068.1 (GeneID 149041).

Additional Product Information

The conjugation of ubiquitin to proteins is an important means to regulate protein activity for many cellular processes by tagging them for degradation. Targeting of proteins by ubiquitination occurs in three main steps that involve the activation of ubiquitin by E1 enzymes, the carrying of ubiquitin to the target protein by E2 enzymes and the ligation of ubiquitin to the target protein by E3 enzymes. Roquin is a member of the RING type E3 ubiquitin ligases. Roquin possesses a CCCH zinc-finger found in RNA-binding proteins and is implicated to play a role in the development of autoimmune disease.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (Cat. No. A120-101P).

Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
