Antibody was affinity purified using an epitope specific to SNF2h/ISWI immobilized on solid support.

The epitope recognized by A301-018A-T maps to a region between residue 1002 and the C-terminus (residue 1052) of human Sucrose nonfermenting protein 2 homolog (SWI/SNF related, matrix associated, actin dependent regulator of chromatin, subfamily a, member 5) using the numbering given in entry NP_003592.2 (GeneID 8467).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

The SWI/SNF-related, matrix-associated, actin-dependent regulators of chromatin (SMARC), also called BRG1-associated factors (BAFs), have been identified as components of the human SWI/SNF-like chromatin-remodeling protein complexes. SNF2h/ISWI, also known as WCRF135, is the gene product of SMARCA5 that associates with WSTF (Williams Syndrome transcription factor) and several other nuclear proteins to mediate chromatin remodeling. SNF2h is also part of the NoRC (nucleolar remodeling complex).

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
