Protein concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.
By immunoelectrophoresis the IgG was shown to 1) react with antiserum specific for bovine IgG and the bovine subclasses IgG1 and IgG2, 2) not react with antiserum specific for IgA or IgM, and 3) produce a single precipitin arc with antiserum against bovine serum identical to that produced with anti-IgG antisera.
By ELISA, the IgG contained less than 1% IgA and IgM. Purified normal bovine IgG is isolated from bovine serum using fractionation and DEAE chromatography.
