Production Procedures
Antibody was affinity purified using an epitope specific to DBC1 immobilized on solid support.

The epitope recognized by A300-433A maps to a region between residues 650 and 700 of human Deleted in Breast Cancer 1 using the numbering given in TrEMBL entry Q6P0Q9 (GeneID 57805).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

The DBC1 (Deleted in Breast Cancer Gene 1 protein) gene is located in a region frequently deleted in breast cancers. In its uncleaved form, DBC1 is localized to the nucleus. Upon cleavage by caspase activity, DBC1 becomes localized to the cytoplasm and functions to promote apoptosis. DBC1 has also been found to bind directly to the estrogen receptor alpha (ERalpha) hormone binding domain to modulate ERalpha expression and mediate hormone-independent breast cancer cell survival.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (Cat. No. A120-101P).
