Antibody was affinity purified using an epitope specific to MAP4K4/HGK immobilized on solid support.

The epitope recognized by A301-503A maps to a region between residue 850 and 900 of human MAPK/ERK kinase kinase kinase 4 (hepatocyte progenitor kinase-like/germinal center kinase-like kinase) using the numbering given in entry AAO32626.1 (GeneID 9448).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.
Antibody was affinity purified using an epitope specific to MAP4K4/HGK immobilized on solid support.

MAP4K4 is a serine/threonine kinase that signals upstream of the c-Jun N-terminal kinase (JNK) and is involved in cell motility. MAP4K4 appears to partially mediate TNF alpha signaling. In skeletal muscle TNF-alpha induces insulin resistance. TNF-alpha induced insulin resistance has been shown to be dependent on MAPK4K4 activity. Interestingly, TNF-alpha regulates MAP4K4 expression and can stimulate MAP4K4 gene expression via TNFR2 (TNFalpha receptor 2) and the c-Jun and ATF2 (activating transcription factor 2) transcription factors. MAP4K4 is highly expressed in most tumor cell lines, and the analysis of MAP4K4 mutants indicates that it may have an important role in cell transformation and invasiveness.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (Cat. No. A120-101P).

Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
