Production Procedures
Antibody was affinity purified using an epitope specific to MED15 immobilized on solid support.

The epitope recognized by A302-422A maps to a region between residue 450 and 500 of human mediator complex subunit 15 using the numbering given in entry NP_001003891.1 (GeneID 51586).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

Additional Product Information

Mediator complex subunit 15 is a component of the Mediator complex, a coactivator involved in the regulated transcription of nearly all RNA polymerase II-dependent genes. Mediator functions as a bridge to convey information from gene-specific regulatory proteins to the basal RNA polymerase II transcription machinery. Mediator is recruited to promoters by direct interactions with regulatory proteins and serves as a scaffold for the assembly of a functional preinitiation complex with RNA polymerase II and the general transcription factors. MED15 is required for cholesterol-dependent gene regulation. It also positively regulates the Nodal signaling pathway. Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (Cat. No. A120-101P).

Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
