Antibody was affinity purified using an epitope specific to p14ARF immobilized on solid support.

The epitope recognized by A300-342A-T maps to a region between residue 82 and the C-terminus (residue 132) of human p14ARF using the numbering given in entry NP_478102.2 (GeneID 1029).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

p16INK4a is a negative regulator of CDK4 and CDK6 kinase activity and cell cycle progression. p16INK4a regulates the cell cycle by inhibiting cyclin/CDK complex formation and the phosphorylation of the retinoblastoma protein (pRb). The gene which encodes p16INK4a, CDKN2A, is frequently mutated or deleted in a wide variety of tumors and is known to be an important tumor suppressor gene. The CDKN2A gene also generates a distinct transcript variant and protein, p14ARF, from an alternate open reading frame (ARF).

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
