Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4. Antibody was affinity purified using an epitope specific to SAPS3 immobilized on solid support.

SAPS3 is a regulatory subunit of the PP6 phosphatase. There are 3 human SAPs (sit4-associated proteins), SAPS1, SAPS2, and SAPS3. The human SAPs are related to SAPs found in yeast that associate with the sit4 phosphatase which is the homolog of human PP6 phosphatase. In yeast sit4 and its associated SAPs are involved in a variety of processes which include transcription, translation, bud formation, glycogen metabolism, ion homeostasis, and telomere function. Because the SAPS3 gene maps to the IDDM4 locus at 11q13, SAPS3 has been implicated in insulin-dependent diabetes mellitus. SAPS domain family member 3, protein phosphatase 6 regulatory subunit 3, PP6R3, and KIAA1558.

Applications
All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
