Antibody was affinity purified using an epitope specific to TMX4 immobilized on solid support.

The epitope recognized by A305-724A maps to a region between residue 299 to 349 of human Thioredoxin-related transmembrane protein 4 using the numbering given in entry Q9H1E5.1 (GeneID 56255).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

TMX4 is a member of the disulfide isomerase (PDI) family of endoplasmic reticulum (ER) proteins that catalyze protein folding and thiol-disulfide interchange reactions. TMX4 has an N-terminal ER-signal sequence, a catalytically active thioredoxin domain, one transmembrane domain and C-terminal ASP/GLU-rich calcium binding domain. Unlike most members of this family, it lacks a C-terminal ER-retention sequence. TMX4 has been shown to have reductase activity in vitro. Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.
